SUNBRIGHT LP4-400GS2 is a degradable PEG derivative incorporating an enzyme-cleavable peptide linker within its structure. It consists of a linear 40 kDa PEG chain (20 kDa x 2) terminated with an NHS carbonate group, allowing efficient conjugation to amine-containing molecules.

The peptide linker is designed to be stable in serum but selectively cleaved inside cells, such as within lysosomes, by enzymatic activity. This cleavage reduces the molecular weight of the PEG, facilitating enhanced clearance from the body compared to non-degradable PEGs. LP4-400GS2 maintains a comparable blood circulation profile to conventional non-degradable PEGs while facilitating faster clearance following intracellular degradation, contributing to improved pharmacokinetic behavior.

An important feature of degradable PEG is the ability to reduce the accumulation of high molecular weight PEGs in the body and to suppress cellular vacuolation, contributing to improved safety profiles in long term therapeutic use.
