SUNBRIGHT BP2-400AL2 is a degradable, 2-arm branched PEG derivative incorporating enzyme-cleavable peptide linkers within its structure. It consists of two 20 kDa PEG chains attached via enzyme-cleavable linkers to a core peptide structure, with an aldehyde group introduced near the branching point for selective conjugation to amine or aminooxy groups on bioactive molecules.
The peptide linkers are designed to be stable in serum but selectively cleaved inside cells, such as within lysosomes, by enzymatic activity. This cleavage reduces the molecular weight of the PEG, facilitating enhanced clearance from the body compared to non-degradable PEGs. BP2-400AL2 maintains a comparable blood circulation profile to conventional non-degradable PEGs while facilitating faster clearance following intracellular degradation, contributing to improved pharmacokinetic behavior.
An important feature of degradable PEG is the ability to reduce the accumulation of high molecular weight PEGs in the body and to suppress cellular vacuolation, contributing to improved safety profiles in long term therapeutic use.
