Performance to suppress non-specific adsorption
BIOLIPIDURE® can effectively suppress the non-specific adsorption of protein on microplate or magnetic beads. Three experiments were carried out to demonstrate the ability of BIOLIPIDURE® to suppress the nonspecific adsorption of protein.
[ By coating microplate ]
- MaxiSorp (Thermo Fisher Scientific) 8-well plate was blocked with10-fold diluted BIOLIPIDURE® (0.5 wt% final, diluted with PBS).
- The plate was aspirated and dried over night at RT.
- HRP-labelled goat anti-mouse IgG was added, and incubated for 1 hr at RT.
- Washing with PBS-T.
- Colorimetric substrate (TMB) was added, and reacted for 10 min at RT.
- Stop solution (sulfuric acid) was added, followed by measuring Abs. at 450 nm.
[ By coating magnetic beads ]
- 8.5 µg of magnetic beads (MagnosphereTM MX100, JSR) was suspended in 100 µL of TBS containing 10-fold diluted BIOLIPIDURE® (0.5 wt% final).
- After 1 hr of incubation at RT, the beads was washed with TBS.
- TBS containing 10 ng of AP-labelled goat anti-human IgG was added, and incubated 1 hr at RT, followed by washing with TBS-T.
- Chemiluminescent substrate (CDP-Star, Roche) was added.
[ Application to Sandwich CLEIA, Microplate-based assay ]
- Mouse anti-human IgG monoclonal antibody was immobilized on MaxiSorp(TM) 8-well plate (Thermo Fisher Scientific).
- The plate was washed with TBS.
- 10-fold diluted BIOLIPIDURE® (0.5 wt%, diluted with TBS) was added, followed by 1 hr incubation at R T.
- Human IgG standard was added, followed by 1 hr incubation at RT.
- The plate was washed with TBS-T.
- AP-labelled goat anti-human IgG polyclonal antibody was added, followed by 1 hr incubation at R T.
- The plate was washed with TBS-T.
- Chemiluminescent substrate (CDP-Star, Roche) was added and reacted for 15 min at RT to measure relative light unit (RLU).
BIOLIPIDURE® significantly enhanced the signal intensity in sandwich CLEIA.
