SUNBRIGHT BP2-400MA2 is a degradable, 2-arm branched PEG derivative incorporating enzyme-cleavable peptide linkers within its structure. It consists of two 20 kDa PEG chains attached via enzyme-cleavable linkers to a core peptide structure, with a maleimide group introduced near the branching point for site-specific conjugation to thiol groups on bioactive molecules (e.g., proteins, peptides, and other biologics).
The peptide linkers are designed to be stable in serum but selectively cleaved inside cells, such as within lysosomes, by enzymatic activity. This cleavage reduces the molecular weight of the PEG, facilitating enhanced clearance from the body compared to non-degradable PEGs. BP2-400MA2 maintains a comparable blood circulation profile to conventional non-degradable PEGs while facilitating faster clearance following intracellular degradation, contributing to improved pharmacokinetic behavior.
An important feature of degradable PEG is the ability to reduce the accumulation of high molecular weight PEGs in the body and to suppress cellular vacuolation, contributing to improved safety profiles in long term therapeutic use.
